Delivery of a spectral shift and microscale thermophoresis device for Biocenter Oulu: for qualitative and quantitative characterization of nanoparticle, including protein, interactions in the range of weak to tight binders in solution without immobilization; assessment of sample quality and aggregation and differentiation of binding events from aggregation; analysis of interactions of fluorescence labelled proteins with small molecules and vesicles; detection independent of the mass ratio of binding partners; fast screening of interactions and sensitive affinity measurement; measurable dynamic range of dissociation constant and binding affinity at least in nM-mM range; low sample consumption (in order of 10 microlitre) and low nanomole of the labeled target required; detection of interaction based on changes of emission spectrum of excited fluorophore under isothermal conditions; creation of a microscopic non-isothermal temperature field (temperature gradient) and determination of nanoparticle, including protein, binding events based on temperature-related intensity change and fluorescence intensity change following thermophoresis under the thermal gradient; measurement and plotting of intensity changes and/or spectrum shifts of the fluorescence probe versus ligand concentrations to calculate the dissociation constant; detailed documentation of all features; integration into the Proteomics and protein analysis core facility infrastructure of Biocenter Oulu; easy to use and maintain instrumentation and software; user interface to control operation; suitable for an academic multi-user environment; maximum budget of 189,000 € (excluding value-added tax); partially funded by European Regional Development Funds (ERDF), BioParts project (A81573).
